It depends on the style of PDF and the kind of Kindle you have. I have an older Kindle DX and I read two-column PDFs on it from time to time. In the end, I find it a bit difficult to read. The reason is that I must read the top half of one column, then go to the next page to read the bottom half, then jump back to the previous page to read the top half of the second column then to the next page to read the bottom of the second column.
While Calibre is great at some document conversion, I found it usually does a poor job converting multi-column PDFs to .azw or .mobi formats (so does everything else I have seen).
It is possible, but for multi-column PDFs I find it much easier to simply read on a computer (though the e-ink on my kindle is much easier on my eyes).
